Can I partition a USB into two and access them both with a copy and paste program even if only one has a letter assigned to it, and copy files that would not appear in Windows?

You're supposed to be asking if you can partition a storage medium accessed with a USB interface. (If this is not the question, then it makes no sense to me.)

The media can be a conventional hard disk, a thumb drive, an SSD...

Of course it can, just as if it were a media running through a SCSI, IDE, EIDE or any other interface.

It is common practice for some manufacturers to include a partition marked as read-only to accommodate add-on software on the USB stick. I would say that with the right knowledge it is no problem to create multiple partitions on any USB stick. I'm not so sure about leaving a partition un-lettered, but I'd have to try that.

At first glance, I don't see any possibility for USB partitioning. You can simply copy and paste using any program that allows you to save the copy.

I think you can't partition to USB, because of the space limitation you have. But you can save files that you don't want to appear on the Windows hard drive.
